Abstract(s)
O presente relatório insere-se na unidade curricular “Estágio” do Mestrado Integrado em
Ciências Farmacêuticas e está dividida em dois capítulos referentes ao trabalho de
investigação no âmbito da descoberta, possíveis utilizações e abordagem toxicológica de
BPC157 e ao estágio em Farmácia Comunitária.
No primeiro capítulo, contempla toda a informação reunida relativa à descoberta,
desenvolvimento, aplicações terapêuticas e abordagem toxicológicas do BPC157. O
BPC157 é um pentadecapéptido, estável no suco gástrico, constituído por 15
aminoácidos. É considerado um mediador da citoproteção/citoproteção
adaptativa/organoproteção, resistindo ao stress provocado por vários agentes nocivos.
Tem interação com várias moléculas, como a serotonina, dopamina e o óxido nítrico. Os
seus efeitos manifestam-se em vários sistemas, contribuindo assim para a homeostase.
O seu efeito pró-angiogénico reflete-se na promoção da cicatrização de vários tecidos,
através da interação com os fatores de crescimento endotelial. Atua no trato
gastrointestinal, mantendo a integridade gástrica. Neutraliza os efeitos tóxicos dos antiinflamatórios não esteroides e do álcool, no epitélio gástrico e noutros epitélios. Combate
a letalidade de certos distúrbios cardiovasculares. Apresenta um efeito cardioprotetor e
neuroprotetor. É usado de forma profilática e terapêutica, em lesões, revertendo a
caquexia associada ao cancro, bem como, a perda muscular e ainda diminui a inflamação.
É no âmbito desta matéria que surge a seguinte revisão, com o objetivo claro de recolher
a máxima informação acerca do BPC157, esclarecer os seus mecanismos e avaliar o seu
perfil toxicológico. Para tal, foi realizada uma pesquisa nas bases de dados PubMed,
Google Scholar e Europe PMC, de onde se retirou literatura científica sobre esta
molécula. Foi possível concluir que o pentadecapéptido pode ser uma aposta promissora
no tratamento e prevenção de certas patologias e que, até agora, não se verificam
entraves toxicológicos à sua utilização, apresentando um perfil seguro. No entanto, é
necessário a realização de mais estudos, que permitam esclarecer alguns mecanismos de
ação, ainda desconhecidos, e a realização de ensaios clínicos, a fim de avaliar se os
resultados convergem com os extrapolados dos ensaios realizados em animais e em
culturas celulares.
No segundo capítulo, encontram-se descritas as atividades desenvolvidas ao longo do
estágio em Farmácia Comunitária, na Farmácia da Praça, em Guimarães, que decorreu
entre fevereiro e agosto de 2020. Este estágio permitiu solidificar todos os conhecimentos adquiridos ao longo do curso e desenvolver competências relacionadas
com a prática farmacêutica.
The present document is part of the curricular unit “Internship” of the Integrated Master’s Degree in Pharmaceutical Sciences and it is divided into two chapters; one is related to the research work in the scope of the discovery, possible uses and toxicological approach of BPC157, and the other to the internship in Community Pharmacy. In the first chapter, all the information gathered regarding the discovery, development, therapeutic applications and toxicological approach of BPC157 is included. BPC157 is a pentadecapeptide stable in the gastric environment, consisting of 15 amino acids. It is considered a mediator of cytoprotection / adaptive cytoprotection / organoprotection, resisting the stress caused by various harmful agents. It has interaction with several molecules pathways, such as serotonin, dopamine and nitric oxide. Its effects are manifested in several systems, thus contributing to homeostasis. Its pro-angiogenic effect is reflected in promoting the healing of various tissues, through interaction with endothelial growth factors. It acts in the gastrointestinal tract, maintaining gastric integrity. In addition, it neutralizes the toxic effects of non-steroidal anti-inflammatory drugs and alcohol on gastric epithelium and other epitheliums, and decreases the lethality associated to certain cardiovascular disorders. It has cardioprotective and neuroprotective effects. It is used prophylactically and therapeutically in injuries, reversing cancer-associated cachexia, as well as muscle loss, and decreases inflammation. It is within the scope of this matter that the following review appears, with the clear aim of collecting the maximum information about BPC157, clarifying its mechanisms and assessing its toxicological profile. In order to do this, search was conducted in the databases PubMed, Google Scholar and Europe PMC, from which articles were extracted. In summary, this pentadecapeptide can be a promising wager in the treatment and prevention of certain pathologies. Until now, there are no toxicological barriers to its use, presenting a safe profile. However, further studies are needed to clarify some mechanisms of action (still unknown), as well as clinical trials, to assess whether or not the results converge with those extrapolated from tests performed on animals and cell cultures. The second chapter describes the activities developed during the internship in Community Pharmacy, at Farmácia da Praça (Guimarães), which took place between February and August 2020. This internship allowed solidifying all the knowledge acquired during the course and also developing skills related to pharmaceutical practice.
