O presente relatório de estágio para obtenção do grau de Mestre em Ciências
Farmacêuticas foi realizado no âmbito da Unidade Curricular “Estágio” do Mestrado
Integrado em Ciências Farmacêuticas da Universidade da Beira Interior (Covilhã). Este
trabalho encontra-se estruturado em três capítulos, referentes a cada um dos períodos
de aprendizagem nas vertentes de Investigação, Farmácia Hospitalar e Farmácia
Comunitária.
O capítulo um é referente ao projeto de investigação intitulado “Desenvolvimentos de
Micro e Nanoemulsões O/A para administração intranasal de Nestorona em Acidente
Vascular Cerebral” desenvolvido no Centro de Investigação em Ciências da Saúde da
Universidade da Beira Interior. Este trabalho teve como objetivo o desenvolvimento de
emulsões nanométricas para o transporte intranasal de Nestorona, um potencial agente
neuroprotetor, de forma a promover o seu direcionamento ao cérebro. As formulações
foram caracterizadas quanto ao seu diâmetro hidrodinâmico médio, índice de
polidispersão (PDI) e osmolalidade. Adicionalmente, foi avaliada a citotoxicidade das
formulações mais promissoras na linha celular de neurónios N27. Foram obtidas
micro- e nanoemulsões de elevada homogeneidade (PDI < 0,2; ou mesmo < 0,1), de pH
e osmolalidade adequados à via nasal, que permitiram incorporar a concentração alvo
de Nestorona (0,48 mg/mL). Os resultados preliminares relativos à citotoxicidade das
formulações na linha celular N27, indicaram que as nanoemulsões desenvolvidas
estavam associadas a uma toxicidade bastante superior à das microemulsões.
O capítulo dois diz respeito ao estágio em Farmácia Hospitalar, realizado nos Serviços
Farmacêuticos do Centro Hospitalar Universitário Cova da Beira, no período de 1 de
Março a 16 de Abril de 2021, sob a orientação da Dr.ª Maria Olímpia Fonseca. São
descritas as competências adquiridas e funcionamento de cada uma das principais
áreas do hospital: Setor de Aquisição e Logística, Setor de Dose Unitária, Setor de
Ambulatório e Setor de Farmacotecnia.
Por último, o capítulo três refere-se ao estágio em Farmácia Comunitária, realizado na
Farmácia Diamantino, Fundão, no período de 19 de Abril a 9 de Julho de 2021, sob a
orientação da Dr.ª Natália Craveiro. Neste capítulo é apresentado o funcionamento da
farmácia e são descritas as tarefas e atividades desempenhadas durante o estágio e
algumas particularidades alocadas ao enquadramento legal desta componente.
This report to obtain the master’s degree in Pharmaceutical Sciences was carried out within the scope of the “Internship” Curricular Unit of the Integrated Masters in Pharmaceutical Sciences of the University of Beira Interior (Covilhã). This work is structured in three chapters referring to each of the learning periods of Research, Hospital Pharmacy and Community Pharmacy. The chapter one refers to the research project entitled “Development of O/W Micro and Nanoemulsions of Nestorone for intranasal administration in Stroke” conducted at Health Sciences Research Center of University of Beira Interior. This work aimed to develop nanometric emulsions for the intranasal transport of Nestorone, a potential neuroprotective agent, to promote its targeting to the brain. The formulations were characterized for its average hydrodynamic diameter, polydispersity index and osmolality. Additionally, the cytotoxicity of the most promising formulations was evaluated in the N27 neuron cell line. Micro and Nanoemulsions with high homogeneity (PDI < 0,2; or even < 0,1), with adequate pH and osmolality for the nasal route, suitable to incorporate the target Nestorone strength (0,48 mg/mL) were obtained. Preliminary results regarding the cytotoxicity of formulations in the N27 cell line indicated that the developed nanoemulsions were associated with a much higher toxicity than microemulsions. The chapter two reports to the internship in Hospital Pharmacy, which was held at the Pharmaceutical Services of Centro Hospitalar Universitário Cova da Beira, from March 1 to April 16, 2021, under the guidance of Dr.ª Maria Olímpia Fonseca. In this chapter are described the acquired skills and the functioning of each main area of the hospital pharmacy: acquisition and logistics, unitary dose and ambulatory distribution, and cytotoxic and compound medicine preparation. Lastly, the chapter three refers to the internship in Community Pharmacy, which was held at Pharmacy Diamantino, Fundão, from April 19 to July 9, 2021, under the guidance of Dr.ª Natália Craveiro. In this chapter are presented the operations of the pharmacy, as well as the tasks and activities performed during the internship and some particularities allocated to the legal framework of this component.
