Este documento encontra-se subdividido em 2 capítulos distintos. O primeiro capítulo contempla o projeto de investigação desenvolvido na área das Ciências Farmacêuticas. O segundo capítulo, por sua vez, aborda a experiência profissionalizante em Farmácia Comunitária.
Os barbituratos (Capítulo I) representam uma classe de fármacos com capacidade de influenciar a neurotransmissão. Essa característica permitiu a sua utilização em desordens neuropsiquiátricas, tornando-se a primeira classe de fármacos com ação sedativo-hipnótica introduzida no mercado. Estudos posteriores revelaram que os barbituratos estão associados a uma diversidade de outros efeitos farmacológicos. Assim, os avanços tecnológicos aliados à descoberta de novos derivados do ácido barbitúrico e tiobarbitúrico impulsionaram a sua aplicação em novas áreas terapêuticas. Com este trabalho de investigação pretendeu-se a obtenção de novos derivados do ácido tiobarbitúrico, nomeadamente bis-tiobarbituratos com potencial atividade inibitória da enzima Xantina Oxidase. A Xantina Oxidase é a enzima responsável pela conversão das purinas endógenas em ácido úrico, estando a sua atividade, consequentemente, ligada ao surgimento da doença comummente conhecida por gota. Assim, todos os derivados bis-tiobarbituratos preparados foram testados como potenciais inibidores desta enzima, tendo-se concluído que todos têm a capacidade de inibir a Xantina Oxidase, obtendo-se percentagens de inibição a uma concentração de 30 µM, próximas ou até superiores às observadas com o fármaco utilizado como referência, o alopurinol. Futuramente, será necessária a realização de estudos complementares farmacocinéticos e farmacodinâmicos.
O farmacêutico é o profissional de saúde vocacionado para a utilização racional do medicamento, assumindo um papel de destaque em várias atividades de promoção da saúde. Assim, no capítulo II, encontram-se descritos os conhecimentos adquiridos durante o estágio em Farmácia Comunitária. O estágio permitiu o contacto com as diversas áreas da farmácia comunitária e o alargamento dos conhecimentos nas áreas do aconselhamento e cuidados de saúde.
This document is subdivided into two separate chapters. The first chapter contemplates the research project developed in the area of Pharmaceutical Sciences. The second chapter addresses the professional experience in Community Pharmacy. Barbiturates (Chapter I) represent a class of drugs capable of influencing neurotransmission. This characteristic allowed its use in neuropsychiatric disorders, becoming the first class of drugs with sedative and hypnotic action introduced in the market. Further studies have shown that a variety of additional pharmacological effects are associated with barbiturates. Therefore, the technological advances associated with the discovery of new derivatives of barbituric and thiobarbituric acid have boosted its application in new therapeutic areas. This work aimed to obtain new thiobarbituric acid derivatives, namely bis-thiobarbiturates with potential inhibitory activity of the enzyme Xanthine Oxidase. Xanthine Oxidase is the enzyme responsible for the conversion of endogenous purines into uric acid and is therefore linked to the onset of the disease commonly known as gout. Thus, all obtained bis-thiobarbiturate derivatives were tested as potential inhibitors of the enzyme. It was concluded that all have the ability to inhibit Xanthine Oxidase, being observed inhibition percentages at a concentration of 30 µM, near or even better than the drug used as reference, allopurinol. In the future, further pharmacokinetic and pharmacodynamic studies will be required. The pharmacist is the health professional dedicated to the rational use of drugs, assuming a prominent role in several health promotion activities. Thus, in Chapter II the knowledge acquired during the internship in community pharmacy is described. The internship allowed the contact with the different areas of Community Pharmacy and to extension of the knowledge in the areas of counseling and health care.
