Abstract(s)
A presente dissertação encontra-se subdividida em três capítulos que abordam cada uma
das etapas da unidade curricular “Estágio” do Mestrado Integrado em Ciências
Farmacêuticas.
O primeiro capítulo aborda a componente de investigação, desenvolvida no Centro de
Investigação de Ciências da Saúde (CICS), da Universidade da Beira Interior. Este focase na verificação da atividade regenerativa de amostras de mel com extrato de própolis
em fibroblastos humanos da derme, seguido de uma análise de um possível sinergismo
entre as propriedades bioativas de diferentes misturas de mel com extrato de própolis.
Ao longo do capítulo, as características fitoquímicas e biológicas das amostras são
determinadas e descritas e os resultados e conclusões apresentados.
Neste sentido, os compostos fenólicos foram determinados através do método clássico
de Folin-Ciocalteu. A determinação dos flavonoides foi efetuada por um método
colorimétrico que usa o cloreto de alumínio. Procedeu-se de seguida à determinação da
atividade antioxidante por dois métodos com princípios e mecanismos distintos - o
método do DPPH baseado na sequestração de radicais livres e o sistema ßcaroteno/ácido linoleico que se baseia na atividade anti-peroxidação lipídica.
Recorreram-se a dois métodos para avaliar a atividade anti-inflamatória e a viabilidade
celular - um método baseado na desnaturação proteica e o método MTT,
respectivamente. Por fim para avaliar a capacidade cicatrizante foi utilizado um ensaio
de cicatrização - scratch wound healing assay.
O segundo capítulo descreve o estágio em Farmácia Comunitária realizado na Farmácia
Holon Covilhã entre 16 de setembro de 2019 e 29 de novembro de 2019, sob orientação
da Dra. Patrícia Amaral. Neste capítulo são abordadas as atividades realizadas que
contribuíram para a minha experiência durante o estágio, assim como ilustradas as
competências e funções dos farmacêuticos.
Por último o terceiro capítulo refere-se ao estágio em Farmácia Hospitalar realizado no
Centro Hospitalar Universitário Cova da Beira, no período de 4 de dezembro de 2019 a
24 de janeiro de 2020 sob orientação da Dra. Olímpia Fonseca. Neste capítulo são descritas as competências adquiridas assim como o funcionamento dos serviços
farmacêuticos.
This dissertation is subdivided into three chapters that focus on the different stages of professional experience needed to obtain a master’s degree in Pharmaceutical Sciences, based on laboratory research, hospital pharmacy and community pharmacy. The first chapter describes the research project conducted at the Health Sciences Research Centre at University of Beira Interior. This project aims to check the regenerative activity of the honey with propolis extract samples in human dermal fibroblasts, followed by an analysis of a possible synergism between different mixtures of honey and propolis extract. Along the chapter, the phytochemical and biological characteristics of the samples are determined and described and the results and conclusions presented. The phenolic compounds were determined with the Folin-Ciocalteu classic method. The flavonoids determination was made with a colorimetric method which uses aluminium chloride. Then, the antioxidant activity was assessed by two methods with distinct principles and mechanisms - the DPPH method based on the free radical scavenging and the ß-carotene/linoleic acid system based on the lipidic anti-peroxidation activity. Two methods were used to evaluate the anti-inflammatory activity and cellular viability - one based on protein denaturation and a MTT method, respectively. Lastly, a scratch wound healing assay was used to evaluate the healing capacity. The second chapter refers to the internship in Community Pharmacy held at Holon Pharmacy Covilhã from September 16th to November 29th, 2019 under the guidance of Dr. Patrícia Amaral. In this chapter the tasks performed, which added to my professional experience during my internship, are described while showing what the role and skills of a pharmacist are. Lastly, the third chapter refers to the internship at the Hospital Pharmacy which took place at the Centro Hospitalar Universitário Cova da Beira from December 4th, 2019 to January 24th, 2020, under the guidance of Dr. Olímpia Fonseca. This chapter describes the skills acquired as well as the operation of the pharmaceutical services.
