Abstract(s)
O presente relatório foi realizado no âmbito da Unidade Curricular “Estágio” do Mestrado
Integrado em Ciências Farmacêuticas da Universidade da Beira Interior. O relatório é
constituído por 3 capítulos: o primeiro diz respeito à componente de investigação e os
restantes descrevem a experiência profissionalizante na área de Farmácia Comunitária e
Hospitalar.
O capítulo I é referente à componente de investigação levada a cabo no Centro de
Investigação em Ciências da Saúde na Universidade da Beira Interior. O trabalho de
investigação iniciou-se pelo estudo da capacidade de formulações, desenvolvidas com base
em trabalhos anteriores, encapsularem e mediarem a transfeção eficaz de siRNA em células
HeLa. Seguiu-se um estudo das características de diâmetro hidrodinâmico e potencial zeta
de formulações semelhantes às anteriores, tendo em vista o seu uso para fármacos lipofílicos
e promoção da mucoadesão. Avaliou-se posteriormente o perfil citotóxico das
nanoemulsões mais promissoras. Os resultados obtidos demonstram uma necessidade de
otimização da formulação para promover a sua eficácia no encapsulamento de siRNA e no
silenciamento da expressão de genes. No entanto, identificou-se um intervalo de
concentrações de lípido catiónico que não aumentam a citotoxicidade da nanoemulsão e
que apresentam características promissoras para um bom desempenho, se administrada
por via intranasal.
Os capítulos II e III relatam a experiência, os conhecimentos e as aptidões adquiridas na
Farmácia Sant’Ana e no Centro Hospitalar Universitário Cova da Beira, respetivamente.
Estes retratam o papel fundamental do farmacêutico na promoção do bom uso dos
medicamentos e no desempenho das suas funções em dois ambientes distintos, mas
igualmente importantes. O capítulo II foca principalmente o circuito do medicamento e
todas as atividades que nele se inserem: receção, aprovisionamento, armazenamento,
distribuição, dispensa. O capítulo III realça as diversas áreas hospitalares onde um
farmacêutico exerce a sua atividade: setor de Dose Unitária, Ambulatório, Aquisição e
Logística e Farmacotecnia. Esta experiência permitiu-me adquirir novas competências e
conhecer as diferentes responsabilidades, funções por mim desempenhadas em cada área
de atividade farmacêutica.
This report was made within the scope of the curricular unit "Internship" of the Integrated Master in Pharmaceutical Sciences of the University of Beira Interior. The report consists of 3 chapters, where the first chapter concerns the component of scientific research and the others describe the professionalizing experience in the area of Community and Hospital Pharmacy. Chapter I refers to the research work carried out at the Health Sciences Research Center at the University of Beira Interior. The research work began by studying the ability of formulations, developed based in previous work, to encapsulate and mediate effective siRNA transfection in HeLa cells. This was followed by a study of the hydrodynamic diameter characteristics and zeta potential of similar formulations as mentioned above, with a view to their use with lipophilic drugs and promoting mucoadhesion. The cytotoxic profile of the most promising nanoemulsions was subsequently evaluated. The results obtained demonstrate a need for optimization of the formulation to promote its efficacy in siRNA encapsulation and gene expression silencing. However, a range of concentrations was identified that do not increase the cytotoxicity of the nanoemulsion and show promising characteristics for good performance if administered intranasally. Chapters II and III report the experience, knowledge, and skills acquired at Farmácia Sant'Ana and Centro Hospitalar Universitário Cova da Beira, respectively. These portray the fundamental role of the pharmacist in promoting the good use of drugs and performing their functions in two distinct but equally important environments. Chapter II focuses mainly on the drug circuit and all the activities that are part of it: reception, stock management, storage, distribution, and dispensing. Chapter III highlights the different hospital areas where a pharmacist works: Unitary Dose distribution, Ambulatory, Acquisition and Logistics, and Cytotoxic and compound medicine preparation. This experience allowed me to acquire new skills and learn about the different responsibilities, functions performed by me in each area of pharmaceutical activity.
