Abstract(s)
Como cada vez mais as Ciências Farmacêuticas são uma área mais abrangente, com uma
ampla gama de saídas profissionais, decidi rentabilizar o estágio ao máximo, dividindo o meu
tempo em três áreas distintas, adquirindo assim no decorrer destes estágios outros
conhecimentos base, para além dos apreendidos durante a parte curricular do MICF.
Sendo assim, o presente relatório encontra-se dividido em três capítulos subsequentes, nos
quais descrevo a minha experiência profissionalizante deste último ano letivo. O primeiro
capítulo refere-se ao estágio em Farmácia Comunitária realizado na Farmácia Diana em Évora
no qual se descreve o que observei e realizei durante o mesmo como, por exemplo, o
funcionamento interno da farmácia, atendimento ao público e prestação de cuidados de
saúde ao utente, indicação farmacêutica e educação para a saúde. O segundo capítulo referese
ao estágio em Farmácia Hospitalar realizado no Hospital do Espírito Santo de Évora, EPE,
no qual se descreve o que observei e realizei durante o mesmo como, por exemplo,
preparação e controlo de produtos farmacêuticos, distribuição de medicamentos, dispensa de
medicação sujeita a legislação em ambulatório e participação do farmacêutico nas visitas
médicas. O terceiro capítulo refere-se ao estágio em Investigação realizado no Centro de
Investigação em Ciências da Saúde na Faculdade de Ciências da Saúde da Universidade da
Beira Interior o qual tinha como objetivo a síntese de Δ5-esteroides oxidados em C7 e a sua
avaliação como potenciais agentes antioxidantes e antiproliferativos em linhas celulares
cancerígenas hormonodependentes (MCF-7 e LNCaP) e em fibroblastos normais da derme
humana. A atividade antioxidante foi avaliada pelo ensaio do DPPH e a atividade
antiproliferativa foi avaliada pelo ensaio do MTT. Os Δ5-esteroides e os seus 7-oxoderivados
não mostraram capacidade elevada para neutralizar o radical livre DPPH, sendo que nenhum
deles diminuiu a % DPPH remanescente para valores inferiores a 50%. Observou-se, contudo,
que os Δ5-esteroides, neste ensaio, evidenciaram maior capacidade antioxidante que os seus
7-oxoderivados. Em relação aos ensaios de viabilidade celular (ensaio do MTT), os compostos
que se mostraram mais tóxicos para as células foram a diosgenina e o seu derivado oxidado.
Neste estudo verificou-se que os Δ5-esteroides têm maior ação antiproliferativa nos três tipos
de células que os seus 7-oxoderivados.
As more and more Pharmaceutical Sciences is a broader area, with a wide range of career options, I decided to divide my practical training time into three distinct areas, acquiring additional knowledge, in addition to those learned during the course of MICF. Therefore, this report is divided into three subsequent chapters, in which I describe my experience during this past year. The first chapter refers to the practical training held in the Community Pharmacy in Farmácia Diana in Évora, which describes what I observed and performed during the same as, for example, the usual work of the pharmacy, customer service and providing health care to user, pharmaceutical couseling and health education. The second chapter refers to the practical training in Hospital Pharmacy performed at Hospital do Espírito Santo Évora, EPE, which describes what I observed and performed during the same as, for example, preparation and control of pharmaceuticals, drug distribution, dispensing medication subject to legislation and participation of pharmacist outpatient medical visits. The third chapter refers to a research period pergormed at CICS-FCS in Universidade da Beira Interior, which aimed the synthesis of Δ 5 -steroids oxidized at C7 and their evaluation as potential antioxidant and anti-proliferative agents in hormone-dependent cancer (MCF-7 and LNCaP) cell lines and normal human dermal fibroblasts. The antioxidant activity was evaluated by the DPPH assay and the antiproliferative activity was evaluated by the MTT assay. The Δ 5 -steroid compounds and their 7-oxoderivatives did not show great capacity to neutralize the free radical DPPH, none of which reduced the % remaining for DPPH values lesser than 50%. Inspite of this, it was observed that, in this assay, Δ 5 -steroids have better antioxidant properties than their 7-oxoderivatives. In cell viability assays, the most antiproliferative agents were diosgenin and 7-oxodiosgenin. In this study, it was also observed that Δ 5 -steroids have higher antiproliferative activity than their 7-oxoderivatives.
