Abstract(s)
presente documento contempla um Relatório de Estágio em Farmácia Comunitária, um Relatório de Estágio em Farmácia Hospitalar e um trabalho de investigação laboratorial intitulado “A oxidação de esteroides pregnanos na preparação de potenciais compostos quimioterápicos”.
O primeiro capítulo corresponde ao trabalho de investigação laboratorial realizado no Centro de Investigação em Ciências da Saúde da Universidade da Beira Interior. Este trabalho consistiu em estudos de oxidação alílica de esteroides pregnanos e sua potencial aplicação. Para isso, partiu-se de três substratos diferentes - acetato de pregnenolona, acetato de 16-desidropregnenolona e progesterona. Verificou-se, no entanto, que, no caso do acetato de 16-desidropregnenolona, acontecia não só uma oxidação alílica como também uma epoxidação. Foram também realizados estudos mecanísticos com dois inibidores radicalares diferentes, que permitiram inferir acerca do mecanismo radicalar de oxidação alílica. Por fim, fez-se uma avaliação da citotoxicidade de dois dos compostos sintetizados em duas linhas celulares de tumores prostáticos (LNCaP e PC-3) diferentes, com o objetivo de averiguar o potencial quimioterápico destes, verificando-se que os compostos têm potencial para progredir para estudos futuros neste âmbito.
O Estágio Curricular em Farmácia Comunitária que realizei na Farmácia Parente, na Covilhã, teve a duração de 3 meses. A descrição desta experiência profissionalizante, que inclui tanto os termos legais como os termos científicos em que se processa esta atividade, encontra-se no segundo capítulo da presente dissertação, correspondente ao Relatório de Estágio em Farmácia Comunitária.
No terceiro capítulo encontra-se o Relatório de Estágio em Farmácia Hospitalar. Este Estágio Curricular foi realizado em Florença, Itália, na Azienda Ospedaliero-Universitaria Careggi, ao abrigo do Programa Erasmus+. Teve a duração de 3 meses. Neste capítulo é feita uma descrição das atividades profissionais que o farmacêutico hospitalar desempenha de acordo com as Boas Práticas da Farmácia Hospitalar e a legislação em vigor.
The present document is composed of a Community Pharmacy Internship Report, a Hospital Pharmacy Internship Report and a laboratorial investigation work concerning the pregnane steroidal compounds oxidation in the preparation of potentially chemotherapic agents. The first chapter addresses the investigation work, accomplished at University of Beira Interior’s Health Sciences Research Centre. This work consisted on the allylic oxidation studies on steroidal pregnane derivatives and some of its applications. For this, three different substrates were used: pregnenolone acetate, 16-dehydropregnenolone acetate and progesterone. With 16-dehydropregnenolone as the subtrate, not only an allylic oxidation, but also an epoxidation, was observed. Some mechanistic studies involving two different radicalar inhibitors were made, in order to attest the radicalar mechanism of the reaction of allylic oxidation. A biologic cytotoxic evaluation was performed on two prostatic cancer cell lines (LNCaP and PC3), in order to test the chemotherapic effect of two synthetized compounds and it was concluded that these compounds have potencial to progress to future studies on this context. I did my three-month Community Pharmacy Internship at Farmácia Parente, in Covilhã, Portugal. The second chapter of this Dissertation describes this professional experience, including legal and scientific aspects of the community pharmacist line of work. I performed my three-month Hospital Pharmacy Internship at Azienda Ospedaliero-Universitaria Careggi, in Florence, Italy, as an Erasmus+ Programme student. The third chapter characterizes the activities performed by a hospital pharmacist following the Good Practice of Hospital Pharmacy rules and the respecting legislation.
