O período de estágio final no âmbito do plano de estudos do Mestrado Integrado em Ciências
Farmacêuticas englobou duas vertentes: um projeto de investigação intitulado “Estudo da
permeação vaginal de Clotrimazol pelo método de Ussing Chambers e o estágio em Farmácia
Comunitária realizado na Farmácia Costa, em Viseu.
A candidose vulvovaginal (CVV) afeta mais de 75% das mulheres em idade reprodutiva pelo
menos uma vez na vida. O Clotrimazol (CT) é um fármaco de aplicação tópica intravaginal,
amplamente utilizado como tratamento de primeira linha no tratamento da CVV. O objetivo
desta investigação prende-se com o estudo do método de Ussing Chambers para avaliação da
permeação de CT através de epitélio vaginal, utilizando um modelo ex vivo suíno. Estes
modelos têm a capacidade de dar informações de eficácia de compostos tópicos, visto que
têm uma excelente correlação de permeabilidade e de resposta tecidual com o tecido vaginal
humano. O método de Ussing Chambers consiste na recolha do tecido vaginal que é
imediatamente montado como uma folha entre duas meias-câmaras – a recetora e a dadora -
estabelecendo o lado apical e o lado basolateral. Foi introduzida também uma técnica de
corte de tecido vaginal. As alíquotas recolhidas da câmara recetora ao longo do tempo foram
analisadas por HPLC para quantificar as concentrações de Clotrimazol.
O estágio em Farmácia Comunitária foi realizado na Farmácia Costa, em Viseu, entre os dias 4
de Fevereiro e 21 de Junho de 2013. Durante o estágio tive a oportunidade de experienciar as
atividades desenvolvidas pela profissão farmacêutica no âmbito da farmácia de oficina,
integrando uma equipa de trabalho direcionada para o utente.
O contacto com ambas as experiências foi o culminar de um ciclo de estudos que contribuiu
não só para a minha formação integrada como também para enriquecimento pessoal no
sentido de me permitir enfrentar realidades diferentes englobadas na profissão farmacêutica.
The period of the final stage in the curriculum of the MSc in Pharmaceutical Sciences comprised two parts: a research project entitled "Vaginal permeation study of clotrimazole by the method of Ussing chamber" and the internship in Community Pharmacy. Vulvovaginal candidiasis (VVC) affects up to 75% of reproductive age women at least once. Clotrimazole is an intravaginal topical drug, largely used as first line treatment of VVC. The aim of this study was the study of an Ussing Chamber technique for evaluation of drug permeation of vaginal application, using an ex vivo porcine model, since data indicate excellent correlation of permeability and tissue response between human and porcine vagina. Ex vivo models can provide important information regarding the penetration and efficacy of topical compounds. The Ussing chambers technique is a tool in which vaginal tissue is collected and immediately mounted as a flat sheet between two half-chambers, establishing a luminal and a serosal side. Moreover, a slicer technique is introduced. Samples from the serosal side were analyzed by HPLC to determine clotrimazole concentrations. The intership on community pharmacy was conducted between February 4th and June 21st , 2013. During this period I had the chance to experience the activities related to pharmaceutical profession under community pharmacy, integrating in a work team patient focused.
