- Nigredo, Albedo Rubedo. Duas, três vezes, riscou a lixa. (...)
- Generating Time Optimal Trajectory from Predefined 4D waypoint NetworksPublication . Ahmed, Kawser; Bousson, K.The main purpose of this paper is to develop a trajectory optimization method to generate optimal trajectories that minimize aircraft total trip time between the initial and final waypoint in predefined 4D waypoint networks. In this paper, the 4D waypoint networks only consist of waypoints for climb, cruise and descent approach without the take-off and landing approach phases. The time optimal trajectory is generated for three different lengths of flights (short, medium, and long-haul flight) for two different commercial aircraft and considering zero wind condition. The Results about the presented applications show that by flying a time optimal trajectory, which was found by applying a single source shortest path algorithm (Dijkstra’s algorithm), can lead to the reduction of average travel time by 2.6% with respect to the total trip time.
- Naphthalene amine support for G-quadruplex isolationPublication . Ferreira, João; Santos, Tiago; Pereira, Patrícia; Corvo, Marta C.; Queiroz, João; Sousa, Fani; Cruz, CarlaG-quadruplex (G4) is involved in many biological processes, such as telomere function, gene expression and DNA replication. The selective isolation of G4 using affinity ligands that bind tightly and selectively is a valuable strategy for discovering new G4 binders for the separation of G4 from duplexes or the discrimination of G4 structures. In this work, one affinity chromatographic support was prepared using a naphthalene amine as a G4 binder. The ligand was immobilized on epoxy-activated Sepharose CL-6B using a long spacer arm and was characterized by HR-MAS spectroscopy. The supercoiled (sc) isoform of pVAX1-LacZ and pVAX1-G4 was isolated from a native sample. Also, the recovery and isolation of the plasmid isoforms from Escherichia coli lysate samples were achieved using an ionic gradient with different concentrations of NaCl in 10 mM Tris-HCl (pH 7.4). The retention times of different DNA/single strand sequences that can form G4, such as, c-MYC, c-kit1, c-kit2, tetrameric, telomeric (23AG), thrombin aptamer (TBA) and 58Sγ3 in this support were evaluated. Our experimental results suggest that the support exhibits selectivity for parallel c-MYC and c-kit1 G4s. In vitro transcription was performed using purified sc pVAX1-G4 and pPH600 to induce G4 formation and circular dichroism (CD) analysis confirmed that both transcripts adopt a parallel G4 topology.
- Surface modification of an intraocular lens material by plasma-assisted grafting with 2-hydroxyethyl methacrylate (HEMA), for controlled release of moxifloxacinPublication . Vieira, Ana Paula; Pimenta, Andreia; Silva, Diana; Gil, Maria; Alves, Patrícia; Coimbra, Patrícia; Mata, Jose; Bozukova, Dimitriya; Correia, Tiago R.; Correia, Ilídio Joaquim Sobreira; Serro, Ana Paula; Guiomar, AntónioEndophthalmitis, an inflammation of the eye due to perioperative infection, may occur after cataract surgery. Intraocular lenses (IOLs) loaded with an antibiotic have been proposed as an alternative to the conventional postoperative endophthalmitis prophylaxis, since the antibiotic is delivered directly to the target site. In this work, an IOL-based antibiotic releasing system was prepared from a copolymer used in the production of IOLs and a fluoroquinolone used in endophthalmitis prophylaxis (moxifloxacin, MFX). Argon plasma-assisted grafting with 2-hydroxyethyl methacrylate (HEMA) in the presence of MFX was the approach selected for surface modification, with MFX loaded both by entrapment in the grafted polyHEMA coating and by soaking. Surface and bulk properties were evaluated before and after surface modification and the MFX release profiles were obtained both in batch mode (sink conditions) and under hydrodynamic conditions, employing a purpose-built microfluidic cell, which simulated the hydrodynamic conditions around the eye lens. The effect of storage on the release profile of the best system was also assessed. The best system released MFX for ca. 15 days above the minimum inhibitory concentration for Staphylococcus aureus and Staphylococcus epidermidis. The released MFX showed antimicrobial activity against these bacteria and was non-cytotoxic against corneal endothelial cells.
- Topics of Measure Theory on Infinite Dimensional SpacesPublication . Velhinho, JoséThis short review is devoted to measures on infinite dimensional spaces. We start by discussing product measures and projective techniques. Special attention is paid to measures on linear spaces, and in particular to Gaussian measures. Transformation properties of measures are considered, as well as fundamental results concerning the support of the measure.
